Irisin, a skeletal muscle-secreted myokine, is produced in response to physical exercise. The glycosylated type I membrane protein is released into the bloodstream by cleavage of the fibronectin type III domain-containing protein 5 (FNDC5) triggered by muscle contraction. It contains a N-terminal signal peptide (amino acid (aa) 1-28), a FNIII domain (aa 33-124), a transmembrane domain (aa 150 -170), and a cytoplasmic tail (aa 171-209). The secreted form of FNDC5 contains 112 amino acids (aa 29-140), and a molecular weight of 12 kDa amed irisin. The expression of FNDC5 is under the control of the peroxisome proliferator-activated receptor γ coactivator 1-α, a master regulator of genes involved in metabolism. Irisin is not only present in skeletal muscle, but also in the cardiac muscle, brain, and skin and in smaller amounts in the liver, pancreas, and other tissues. Irisin also plays an important role in central nervous system (CNS) diseases, such as neurodegenerative diseases, cerebral ischaemia and Alzheimer’s disease.

This species-specific MonELISA® Irisin (FNDC5) ELISA is a colorimetric based sandwich immunoassay utilizing an antibody to the specie’s Irisin (FNDC5) bound on the surface of microwells as the capture antibody and a biotinylated antibody to Irisin (FNDC5) as the detection antibody. When antigen is added to the well it is bound by the immobilized antibody. The biotinylated antibody then binds to the antigen forming an antigen-antibody complex. Streptavidin-Peroxidase in the presence of an enzyme substrate quantifies analyte bound. Color development is directly proportional to the antigen concentration in the sample.
