Recombinant human HtrA4 (amino acids 138 – 476, BC057765) is expressed in E coli. with a C-terminal His-tag. The calculated Mr of HtrA4 is 37,6 kDa. HtrA4 is solubilized in 120 mM NaCl, 120 mM Imidazol, 15% (v/v) Glycerol, 40 mM Tris-HCl, pH 7.5.

Overview: 

HtrA4 is a secreted human serine protease is supposed to form oligomers of identical protein chains as revealed by X-ray analysis for the bacterial HtrA protein DegP [1]. Polypeptide chains of human HtrA4 consist of several domains: An N-terminal insulin-like growth factor domain is followed by a Kazal-type serine protease inhibitor domain, a linker region, a trypsin-like protease domain and a PDZ domain [2]. The truncated version HtrA4D covers part of the Kazal domain, the serine protease and the PDZ domain. The function of HtrA4 is a probable serine protease. No information is available yet on physiological substrates or interacting proteins.

Purity: Recombinant HtrA4 appears as a major protein of about 38 kDa in SDS-PAGE (> 90% of total protein). Due to minor N-terminal truncation of the protein it may appear as a double band.

 

Enzymatic Activity: Proteolytic activity of recombinant human HtrA4 is documented by digestion of ß-casein.  0.5 mg ß-casein/ml are completely digested by 10 μg/ml HtrA4 within 24 hours at 37 °C (see PDF insert).

 

Stability & Storage: Recombinant human HtrA4 is stable until the expiry date given on the label when stored at -70 °C. Repeated freezing and thawing should be avoided. Note: Upon freezing, a faint precipitate may form which contains HtrA4. This precipitate was shown to contain catalytically active HtrA4 in a casein digest. Therefore, we recommend vortexing the protein solution before use.

 

Applications: Recombinant HtrA4 allow detailed studies of the structure and function of this protease. The enzyme is used to screen for inhibitors and to characterize inhibitor actions. Recombinant HtrA4 can also serve as standard in enzymatic and immunochemical assays.
