Platelet-derived growth factor (PDGF) is a dimeric glycoprotein composed of two disulfide-stabilized polypeptide chains, A and B, forming heterodimers (PDGF-AB) or homodimers (PDGF-AA and PDGF-BB). PDGF signals through receptor tyrosine kinases PDGFRα andPDGFRβ and activates pathways involving MEK/ERK, EGFR, Src, and PI3K/AKT. It is a potent mitogen for mesenchymal cells such as fibroblasts, glial cells, and vascular smooth muscle cells. It is implicated in atherosclerosis, glomerulonephritis, cancer, and vascular smooth muscle cell contraction. PDGF-AA regulates VEGF expression in non-small cell lung carcinomas and mediates the proliferation and differentiation of oligodendrocyte progenitor cells. PDGF-AA is also used to differentiate human pluripotent stem cell-derived neural progenitor cells into oligodendrocyte precursor cells.
